- Bebop (Cam Clarke)
- Donatello (Oliver Vacquer)
- Leatherhead (Andre Soggliuzzo)
- Leonardo (Cam Clarke)
- Michelangelo (Peter Oldring)
- Raphael (Roy Samuelson)
- Rocksteady (Andre Soggliuzzo)
- Splinter (Danny Woodburn)
Notes
Cam Clarke voiced Leonardo in the 1987 TMNT series as well as Rocksteady but in this game he voices Leonardo and instead, Bebop who was originally voiced by Barry Gordon who also voiced Donatello. Danny Woodburn performed the motion capture for Splinter in the movie and was originally meant to voice him but was dubbed over by Tony Shalhoub in the finished film however Woodburn does actually voice Splinter in the game. The Android and IOS version of the game does not feature the Shredder. Shredder does, however appear in the 3DS version. The main screen theme is an instrumental remix of the original 1987 theme.
Nintendo 3DS cast
- April O'Neil (Julie Nathanson)
- Baxter Stockman (K. Todd Freeman)
- Donatello (Oliver Vacquer)
- Leonardo (Cam Clarke)
- Michelangelo (Peter Oldring)
- Raphael (Roy Samuelson)
- Rat King (Jason Spisak)
- Shredder (William Fichtner)

Notes
In early production, William Fichtner was meant to portray the Shredder in the film, however, he was replaced by Tohoru Masamune in later development and his character, Eric Sacks wasn't the Shredder although he was meant to be. Fichtner still voiced the Shredder in this game.
